What Is AOD-9604?

AOD-9604 is a modified fragment of human growth hormone (hGH 177-191) that was developed to isolate the fat-burning properties of GH without its growth-promoting or insulin-desensitizing effects. It completed Phase 3 clinical trials for obesity in Australia but did not receive regulatory approval. Researchers study it as a lipolytic compound that activates beta-adrenergic pathways and inhibits lipogenesis.

Research Observations - Not Medical Claims
  • Phase 3 obesity trial (METAOD study): AOD-9604 did not achieve statistically significant weight loss vs. placebo in the primary endpoint of the Phase 3 trial - the main reason it was not approved for obesity (PMID: 16413786).
  • Lipolysis mechanism: AOD-9604 mimics the lipolytic action of hGH - it activates beta-3-adrenergic receptors in adipose tissue and inhibits acetyl-CoA carboxylase, the rate-limiting enzyme in de novo lipogenesis (PMID: 11600718).
  • No IGF-1 effect: Unlike full-length GH, AOD-9604 does not stimulate IGF-1 production or promote anabolic tissue growth - important safety differentiation in long-term use.
  • No glucose disturbance: Rodent studies confirmed AOD-9604 does not impair glucose tolerance or insulin sensitivity - a key advantage over full-length GH.
  • Cartilage repair: Surprising finding - AOD-9604 showed regenerative effects on cartilage in in vitro and animal models; this has driven interest as a potential OA treatment (PMID: 24403479).
